Реферат: We generated novel, effective candidate vaccine against Brucella abortus based on recombinant influenza viruses expressing the Brucella ribosomal protein L7/L12 or outer membrane protein (Omp)-16 from the NS1 open reading frame. The main purpose of this work was to evaluate the safety, immunogenicity and protectiveness of vaccine candidate in laboratory animals.

Реферат: The paper presents the results of a study of cultural characteristics of the strain A/equine/Otar/764/07 (H3N8) of equine influenza virus isolated from sick horse during the outbreak of equine influenza epizooty in the Republic of Kazakhstan. The studies determinedthe optimal culture conditions for equine influenza virus strain A/equine/Otar/764/07 (H3N8) in MDCK cell culture. The optimal infecting dose of the virus was 0,1 TCE50/cell, temperature and cultivation duration - 34 + 0,5 әC, 72 h, respectively, and Trypsin concentration in medium - 2 mg/ml. Subject to the determined parameters of A/equine/Otar/764/07 (H3N8) strain cultivation in MDCK the virus - containing suspension with an infectious titer of at least 7,00 lgTCE50/ml and hemagglutinating titer of 8,0 log2 can be obtained. The results obtained during the strain reproductive characteristics study can be used for the development of vaccines and diagnostic kits.

As a result of the conducted laboratory researches of the delivered biomaterials from sick, died goats and sheep from various regions of the Republic of Kazakhstan, Tajikistan and Kyrgyzstan it has been demonstrated that virus of this disease is present. Реферат: The paper presents cultural characterization of Peste Des Petites Ruminants (PPR) virus and sheep pox virus in co-culture. During the study we selected optimal cell cultures such as lamb kidney cells (LK), lamb testicles cells (LT) and goat gonad cell line (CG-91) allowing to obtain virus suspension with high infective activity. The optimum period of cultivation is 7 days and the infecting dose of PPR and sheep pox viruses is 0,01 TCID50/cell. The interference phenomenon absence between the two viruses is confirmed by electron microscopic studies, as well as the cytopathic effect of these viruses in cell culture. Subject to the above parameters highly active virus-containing suspension can be obtained for PPR and sheep pox associated vaccine production.

Реферат: The article presents the results of phylogenetic analysis of nucleotide sequences of Camelpox virus strain M-96 isolated in the Mangistau region, Republic of Kazakhstan. The nucleotide sequences of 7 genes of twenty-six CPV strains / isolates were selected from the international GenBank database to determine the degree of M-96 strain nucleotide sequences homology with the other CMLV strains using phylogenetic analysis. As a result of the conducted comparative phylogenetic analysis we determined genetic differences and the degree of Kazakhstan strain homology with other CMLV strains, isolated at different times and in different geographical regions and the ways of possible CMLV introduction on the territory of the Republic of Kazakhstan. Kazakhstan strain M-96 of Camelpox virus showed 100% homology with five genes of CMS strain (Iran, 1970), two genes of SP-1_Iran strain and one gene of Bikaner strain, isolated in 2008 in India. The obtained data analysis suggests that Camelpox virus can be transmitted to the territory of the Republic of Kazakhstan from Iran via Turkmenistan.

Реферат: As a result of extensive research, the highest immunological parameters were obtained in the groups of animals immunized with the mixture of designs NS1-124-L7L12 + NS1-124-Omp 16 (H5N1 and H1N1), not inferior piece of the vaccine. RYR2 sequencing reveals novel missense mutations in a Kazakh idiopathic ventricular tachycardia study cohort
Реферат: Channelopathies, caused by disturbed potassium or calcium ion management in cardiac myocytes are a major cause of heart failure and sudden cardiac death worldwide. The human ryanodine receptor 2 (RYR2) is one of the key players tightly regulating calcium efflux from the sarcoplasmic reticulum to the cytosol and found frequently mutated (<60%) in context of catecholaminergic polymorphic ventricular tachycardia (CPVT1). We tested 35 Kazakhstani patients with episodes of ventricular arrhythmia, two of those with classical CPVT characteristics and 33 patients with monomorphic idiopathic ventricular arrhythmia, for variants in the hot-spot regions of the RYR2 gene. This approach revealed two variants; one de-novo RYR2 mutation (c13892A>T; p.D4631V) in a CPVT patient and a novel rare variant (c5428G>C; p.V1810L) of uncertain significance in a patient with VT of idiopathic origin which we suggest represents a low-penetrance or susceptibility variant. In addition we identified a known variant previously associated with arrhythmogenic right ventricular dysplasia type2 (ARVD2). Combining sets of prediction scores and reference databases appeared fundamental to predict the pathogenic potential of novel and rare missense variants in populations where genotype data are rare.
